Since you can't beat that Initiative roll, I'll post my first turn.
Shooter McArcher, Turn 1
Start in Y-9 holding bow, potion in belt.
Move: Move to V-10
Standard: Ready an action
Shoot Boris when I have Line of Effect
End of turn.
Stats:

AC: Full 16, FF 12, Touch 14

surprisingly you go first (not really, slow dwarf ;-) )
Boris starts in b12 psionicly focused woth ranseur in hand

Against normal vision shadow illumination gives anyone within basic concealment (20% miss chance) and allows hiding but does not automatically make a character hidden.
LoS existed between Shooter (V-5) and Boris (B-19), but LoS is broken by the pillar as Borus moves east out of G-19.

Well, since I had LOS and LOE when I readied my action, does it go off immediately? If my readied action would not go off before my turn, can I then revise my turn to just take a shot at him since I wasn't aware that I had LOS?

Ok, I'm confused. Do you measure LoS from the centre of their square, or from any part of it? Some refs seems to do it differently.

LoS is measured from corner to corner as shown in a diagram on page 139 of the Player Hand Book. I think there's also a text description in the SRD but I can't find that at the moment.

I'd forgotten about the readied action...
Since LoE exists between V-5 and B-19, your readied action would instantly trigger. You may rewind to take the shot if you wish.
Note that Boris does have the benefit of both cover and concealment.

Ok, then in light of having LOS after his move, Shooter's readied action will change to be to shoot at Boris when he has LoE and Boris does not have cover. I'm assuming that goes off when he moves out from behind the pillar, so I'll make my attack.
Shooter's readied action changed in the spoiler above in light of having LOS after his move to V-5. It goes off when Boris no longer has cover relative to Shooter, probably in C-19 or D-19.

It's generally best to post LOS requests in the Waiting Room. The refs occasionally peruse the current games, but they may miss this one. I've gone ahead and made the request for you.

Keep in mind that you have cover from me. It hasn't mattered yet since all of my hits have either been before you were there or hit anyway. Also, since I'm now in the shadowy illumination, you need to roll concealment for attacking me too.

Cover is a bonus to AC, not a penalty to attack. And the darkvision extends 60ft from your character.
