if magic missile is enhanced with fell drain and multiple missiles are directed at the same person, do they gain multiple negative levels?

No, because it grants only 1 negative level to the spell. But if directed all over the place it could be quite effective

Yup.
Maximum of 1 negative level per target. You can get a billion targets if you use the right spell though.
