If i cast spell with metamagic rod, i need two hands?

One hand for somatic, and one hand for holding rod.

or i need one hand for somatic and holding rod?

and if i need two hands, so i lose ac bonus from buckler?

or i just need to hold the metamagic rod?

You would need two hands, yes. One to wield the rod and the other for somatic components.

Emphasis mine. The metamagic rods specifically stat the rod must be wielded to be used.
