Hello, glad to hear someone else is stepping up to run a game on this wonderful site. I suggest you read this (http://www.giantitp.com/forums/showthread.php?t=257924) first so you get an idea of how to run a pbp game on this forward. On the subject of content it really depends on the edition you want to play and whether or not you want to use a module or homebrewed setting. If you're asking for idea's for a homebrewed setting it is somewhat redundant since it completely relies on the DM's imagination. I suggest you run one of the many modules available for the edition you wish to run until you are more comfortable or inspired to run a homebrewed setting of your own.
