What do you all think would be a good class/classes progression for maximizing the abilities of the Mother Cyst feat? ...involving undead flavor would be ideal, I suppose.

Really the mother cyst chain doesn't need to be the main focus of a build. Since it's so hard to get the tumor on creatures easily in combat. Apply a 2nd level touch spell isn't exactly fun.

I generally build mother cyst builds in 2 ways.
1: Try to get the dominate + necrotic tumor spells are early as possible usually abusing the versitile spellcaster feat with a class that knows its entire spell list so you can spend 2 spell slots to cast a level higher than normal such as dread necromancer. (Note some people don't think versatile spellcaster works that way so check with your DM)

2: Try and build a character that can knock out enemies without killing them so you can implant the cysts in them later. Something like beguiler or just a standard wizard.

Most of the value of Mother Cyst comes out of combat. Implanting a cyst in someone just to blow them up a round later is a waste of time and spell slots.
Specifically in that you can essentially permanently dominate anyone you can get your hands on. All the other spells are just gravy, but not worth a feat.

Not only is it a (non-mind-affecting) Necromancy effect, it's also really hard to detect and remove, so it's an ideal option if you want to covertly infiltrate an organization and take over with your minions.
Of course you can also use it to build yourself an army of minions, but that's a good way to slow combat to a crawl and get books thrown at your head.

Silva has pretty much covered the main points. Optimizing it just requires getting the necessary spells asap and increasing your save DCs.
What class or build you use is irrelevant beyond that, at least with respect to the effectiveness of your cysts. A Rod of Reach Spell can be nice, but it's not really necessary because knocking people out isn't that hard.

Throw it on a Social focused Enchanter, throw invisible/still/silent/heightened necrotic cysts on everyone you meet via handshakes pats on the back friendly hugs etc.

Stealing an idea from PiggyKnowles, Duskblade 3 lets you channel the spell with a melee attack, then use Mosquito's bite to make enemies not notice you hit them. If you conceal that you are attacking, then they won't even notice you attacked. Keep hidden, sneak up, then move on.
