"Special business" refers to Sora's relationship with Kairi.

"It" and "Little secrets" refers to what Riku thinks Sora is talking about.

"I knew it! Cheater!" is exclaiming Kairi's viewpoint on the conversation. She thinks that Sora is talking about an affair.

Sora and Riku aren't gay. Sora has a relationship with Kairi. The joke is that, based on what Sora and Riku said, Kairi made the conclusion that Sora was cheating on her. That idea was brought on by "little secrets." The sub-joke was that Riku is so confident that he couldn't have caused Kairi to yell.

Is it still confusing?

Kairi: The only girl, in the upper left corner. Has a relationship with Sora (See above). Thinks she is being cheated on by Sora.
