Can a creature simultaneously have an Item Familiar (https://www.d20srd.org/srd/variant/magic/itemFamiliars.htm) and a true Wizard familiar like a rat? Where is the rules text for this?

I don’t have text for this but they seem to be entirely separate mechanics, since you have to be a caster to have a traditional familiar but any schmuck can get an Item Familiar.

yup, the two are different things, you can have both, plus animal companion and wild cohort too.
there is nothing that says one replaces the other.

If you had a way to take a construct creature as a familiar, you could also make it an item familiar. An Improved Familiar homunculus, maybe?

Also, deities can make any creature into a familiar, so a god could take, say, an adamantine golem, make it his familiar, then make it his item familiar (and thus granting it sentience). Alternately, make it intelligent via Item Familiar, then make it his regular familiar.

There are Weapon Familiars for Warforged in Eberron, but I'm pretty sure you have to swap out your normal familiar to get them. Same for the Imbued Staff out of Dragon magazine, which is kind of similar.
