Arcane Disciple + Spell Mastery + Signature Spell
Let's say a wizard (or other arcane prepared caster) takes the Arcane Disciple feat and is able to learn the Heal spell through the Healing domain. The normal restriction, since he is a prepared caster, if that he may only prepare it once. If he then takes Spell Mastery and then Signature Spell (Heal), is he able to cast Heal more than once a day then since he technically isn't preparing the Heal spell more than once?

If you cast something spontaneously, that isn't preparing it.
This trick works by RAW as you described.

Like said above, it does not work.
You could probably make it an Archmage SLA, though.
