It creates an overflow error and starts you over at assassin 1.

Here is the relevant rule, found on page 206 of the DMG.

Despite the twenty-level limit indicated in the Player’s Handbook, you can advance a class level beyond 20th by using the rules in this book. You can also advance the class level of a ten-level prestige class beyond 10th level, but only if the character level of the advancing character is already 20th or higher. You cannot advance the class level of a class with fewer than ten levels beyond the maximum described for that class, regardless of the character level of the advancing character.

This is the only general rule for this situation that I'm aware of. Based on this, since you cannot improve the casting any further, you are forced to choose a different class to advance. If it's too late—for example, if you're a Rogue 5/Assassin 5/Unseen Seer 5 and you take another level of assassin—the spell advancement simply has no effect, as the epic progression for it is unavailable.

A 136 Yes. From Complete Arcane : "A requirement based on a spell measures whether the character [...] is capable of producing the necessary effect". The rest of the text eplicitly mentions invocations and SLAs‚ but (Su) ability that explicitly reproduce a spell are "producing the necessary effect".

It depends on the exact wording of the prerequisite, but in most cases, no. Su abilities aren't cast and don't count as spells, so they can't fulfill a requirement that you be able to cast a particular spell.

There's not really much to say about it. The necessary effect is the ability to cast magic missile as a spell, something that a Su version of magic missile does not provide, since it is not a spell. SLAs work because there is a specific exception explicitly laid out for them in the rules. That's not the case here.

The setting information states that it's essentially a dungeon at the end of which is a magical meteor which grants godhood. I think there's a module for it, but I'm not 100% sure.

It looks like the prices are multiplied tenfold in the ELH from the +6 mark.
I assumed this +6 meant "+6 armor bonus" at first, but since you can also have something like :

+10 total armor bonus (+5 armor and +5 various bonuses) it... does not make sense...

The +1 enhancement bonus is not on you, it's on the leather armor. It enhances the item's armor bonus by 1. The +1 leather armor provides 3 armor bonus, which does not stack with the 4 armor bonus provided by mage armor.
